Dr. Holtorf's Immune Modulation Protocol

Three peptides for immune modulation, gut healing, and telomere maintenance. TA-1 at clinical dose (1.6mg 2x/week), oral BPC-157 for gut health, and Epithalon in 10-day pulses. Note: Holtorf co-founded Integrative Peptides which sells oral BPC-157.

Healing & Recovery

For immune modulation, gut healing, and telomere maintenance using three peptides at clinical doses.

What This Protocol Does

TA-1 at 1.6mg twice weekly provides ongoing immune modulation through T-cell activation. Oral BPC-157 at 500mcg daily targets GI healing directly through the gut lining. Epithalon runs as a 10-day pulse every 6 months for telomerase activation and telomere maintenance. Each sub-stack is standalone and targets a different system. The oral BPC-157 route is unusual; most protocols use subcutaneous injection.

Who Should Use This

Intermediate users looking for immune support, gut healing, or longevity in a simpler package than a 4-5 peptide stack. Disclosure: Holtorf co-founded Integrative Peptides LLC, which sells oral BPC-157. The FTC issued a warning letter for unsubstantiated claims. Evaluate the protocol on its merits, but factor in the commercial interest. Budget is moderate since TA-1 and oral BPC-157 run long-term while epithalon is only pulsed twice per year.

What the Research Says

Thymosin alpha-1 is approved in 30+ countries and has clinical trial data showing improved T-cell counts, NK cell activity, and dendritic cell maturation (Tuthill et al., PMID 10813511). The standard clinical dose (1.6mg) matches the Zadaxin label used in hepatitis trials. BPC-157 has 100+ preclinical studies on tissue repair and gut protection (Sikiric et al., PMID 9073154). Oral bioavailability data for BPC-157 is limited; most published studies used injection. Epithalon follows the Khavinson protocol for telomerase activation (Khavinson, PMID 14523363). No studies test this 3-peptide combination together.

Wolverine Stack

BPC-157 + TB-500 + Ipamorelin +2

Five peptides, three sub-stacks, and a torn hamstring that started it all. Ben Greenfield published full dosing for BPC-157, TB-500, ipamorelin, tesamorelin, and GHK-Cu after using the combo to recover from a persistent upper leg injury. The healing duo (BPC-157 + TB-500) can be run alone; the GH pair (ipamorelin + tesamorelin) adds recovery optimization at bedtime. Budget $400-800/month and plan for 4-12 weeks on, equal time off.

Never Get Sick Stack

TB-500 + Thymalin + LL-37

Three peptides, three different cycle lengths, one goal: stop getting sick. Greenfield pairs TB-500 (3 months on, 3 off) with Thymalin (3-10 day bursts every 6-12 months) and LL-37 (4-6 week antimicrobial cycles). Each targets a different layer of immune defense. Warning on LL-37: Herxheimer reactions are common in the first two weeks if you have SIBO or gut dysbiosis.

Dr. Koniver's GH Secretagogue Bedtime Stack

BPC-157 + Ipamorelin + Tesamorelin

One syringe, three peptides, taken at bedtime. Dr. Koniver confirmed on Huberman Lab Ep 197 that he combines BPC-157 (500mcg), ipamorelin (100mcg), and tesamorelin (2mg) in a single subcutaneous injection. Schedule is 5 days on, 2 off. No food for 45 minutes before. The single-syringe detail settled a long-running debate about whether these peptides could be mixed safely.

Frequently Asked Questions

Oral bioavailability data for BPC-157 is limited. Most published research used injection. Holtorf advocates oral dosing specifically for GI conditions, arguing the peptide acts locally on gut tissue before systemic absorption. Community reports on oral BPC-157 for gut issues are positive, but the evidence base is weaker than for injectable BPC-157. Holtorf's company sells oral BPC-157, so factor in the commercial interest.

Dr. Holtorf co-founded Integrative Peptides LLC, which manufactures and sells oral BPC-157 products. The FTC sent a warning letter regarding unsubstantiated health claims. This doesn't mean the protocol is wrong, but the financial incentive to recommend oral BPC-157 specifically should be considered when evaluating this protocol versus injectable alternatives.

Greenfield uses TB-500 + thymalin + LL-37 (3 injectable peptides for immune defense). Holtorf uses TA-1 + oral BPC-157 + epithalon (immune + gut + telomeres). Greenfield's targets acute immune defense. Holtorf's is broader, covering gut health and longevity alongside immunity. Greenfield's requires more injection management; Holtorf's includes an oral component.

TA-1 at clinical doses (1.6mg, 2x/week) has been used long-term in hepatitis treatment protocols. The Zadaxin label supports ongoing use. Holtorf recommends it as a maintenance peptide. Monitor complete blood count periodically. Consult your physician if you're on immunosuppressant medications, as TA-1 stimulates immune activity.
